Embodiments of nanostructured, multilayered metal-dielectric particles suitable for use as Raman spectroscopic probes, as well as methods of designing, making and using such multilayered nanoparticles, and kits including the multilayered nanoparticles. The multilayered nanoparticles include alternating metal and dielectric layers and an outer dielectric shell. One or more of the dielectric layers may include a plurality of reporter molecules. Embodiments of the multilayered nanoparticles are suitable for detecting target analytes in a sample. Some embodiments of the multilayered nanoparticles are suitable for use in multiplexed assays, including assays for multiple target analytes having differing concentrations.

Biosensors based on one- or two-dimensional photonic-crystal reflectance filters operating at near-ultraviolet wavelengths. The biosensors feature a tightly confined resonant electric field at the surface of this biosensor and provide a high surface-sensitivity to bulk-sensitivity ratio, enabling enhanced detection resolution for biomolecules adsorbed on the biosensor surface. These new biosensors can be fabricated in mass by replica molding and are especially well suited for applications requiring the detection of small molecules or ultra-low analyte concentrations.

This technology is a viral vector based on the murine leukemia virus (MLV), which has been favored for clinical gene therapy due to its preferential infectivity of human cell types. The original vector has been modified through the use of directed evolution targeting its protease (PR) gene, which has been found to produce increased stability for the viral vector. It is believed that this modification has the potential to be applied to all clinically-relevant viral vectors.

With the use of CRISPR-AID technology targeted genes can be simultaneously upregulated, downregulated, and deleted allowing discovery of synergistic effects to enhance strain phenotypes. Utilizing this technology will allow fast generation of novel strains suited to industrial applications. In addition Dr. Zhao has generated novel CRISPR constructs optimized for CRISPRa and CRISPRi applications for use in yeast. This is the first use of orthogonal CRISPR strains in yeast and will greatly expand the toolbox for yeast genome engineering.
